Marie Maynard Daly was a pioneering scientist, known for her work at the interface of analytical chemistry and biochemistry. Her doctoral research at Columbia Chemistry was centered on the characterization of digestive enzymes and their reaction products. In the spirit of Marie Maynard Daly’s passion and prowess for analytical biochemistry, I will present work conducted by my group at Columbia Chemistry on the development of new analytical methods to characterize cell signaling enzymes.<br/> <br/>Signal transduction through protein tyrosine phosphorylation is critical for many core functions of animal cells, including proliferation, survival, programmed death, and cell-cell communication. These processes are mediated by two large enzyme families, tyrosine kinases and tyrosine phosphatases, that collectively control the phosphorylation states of thousands of sites in human proteins. Our lab is focused on the development of new biochemical and analytical tools to dissect the functions of phosphotyrosine signaling enzymes. We are particularly interested in understanding how the substrate and ligand sequence specificities of tyrosine kinases and phosphatases shape and tune cell signaling pathways.<br/> <br/>In this talk, I will describe a high-throughput biochemical platform that we have developed to profile the sequence specificities of phosphotyrosine signaling enzymes. I will discuss how this technique has enabled the large-scale characterization of disease-associated mutations surrounding tyrosine phosphorylation sites. I will also highlight our efforts to understand the sequence-dependent activation of tyrosine phosphatases that contain regulatory phosphotyrosine-recognition domains. These specificity profiling studies are shedding new light on the molecular basis for sequence recognition by phosphotyrosine signaling enzymes. These studies are also revealing how the sequences of tyrosine phosphorylation sites fine-tune signal transduction.
