Bioorthogonal reactions have found wide applications in chemical biology, including specific labeling biomolecules like lipids, proteins, nucleotides and glycans, efficient generation of antibody-drug conjugates and protein degradation through proteolysis targeting chimera. However, conventional real-time ligations suffered from the lack of spatial and temporal resolution which are equally important in biology. Here, we developed some modular scaffolds with controllable reactivity. These scaffolds, when in caged state, remained inert while reacted promptly with partners when in uncaged state. Also, various scaffolds which can be uncaged by different stimuli, such as light, biomolecules and enzymes, have been developed. These molecules could be used in biomolecules labeling with both high spatial and temporal resolution and high specificity. Proof of concept has been demonstrated in specific organelles imaging in live cells and living animals.
