Warfarin is an important anticoagulant agent. It is synthesized by the conjugate addition of 4-hydroxycoumarin with benzalacetone. Our study describes the use another type of MIchael acceptor - alkenyl-substituted N-heterocycles - in reactions with 4-hydroxycoumarin. We demonstrate that an acid-catalyzed methodology is effective for preparing heterocyclic derivatives as structural analogs to warfarin. In addition to tollerating a variety of substituents on the 4-hydroxycoumarin, the chemistry may be done with several types of N-heterocycles. Moreover, similar chemistry has been done with thiocoumarin and 2(1H)-quinolinone.
