Protein homeostasis is the result of dynamic cellular control of protein concentration, location, and quality. Loss of protein homeostasis is a precursor to disease and death. Multiple insightful studies into control of proteostasis have been done previously using individual reporter constructs. A true understanding of proteostasis can only be achieved if we quantify the synthesis, folding, activity, and degradation of each protein in the proteome as homeostasis varies. Using a simple kinetic model as our framework, as few as four measurements can describe proteostasis control simultaneously for hundreds of proteins in vivo.  I will share some of our current efforts showing that this approach is applied equally well to humans and model systems across the entire proteome.
