4D bioprinting is an intriguing and emerging biomanufacturing technique to fabricate self-assembly structures that can dynamically change their shapes over time. Currently, there are limited biomaterials that are suitable for 4D bioprinting applications. Therefore, the objective of our study is to create novel nano or smart bioinks to fabricate advanced 4D multi-responsive constructs for complex tissue regeneration (such as cardiovascular tissue and neural tissue). In our recent study, a 4D programmable culture substrate with a self-morphing capability has been created to fully reflect the supportive biological environment in which stem cells reside in vivo. It was found that a time-dependent 4D process plays an essential role in regulating stem cell behaviors in a spatiotemporal manner, and enhances in vitro differentiation of stem cells. Furthermore, a novel 4D hydrogel-based cardiac patch with physiological adaptability has been printed in our recent breakthrough work for myocardial infarction treatment. Results demonstrated that the printed patch could transform from a 3D flat pattern to a curved architecture when appropriate printing parameters are selected. This unique 4D self-morphing capacity has been shown to improve both the biomechanical properties of the patches themselves and the dynamic integration of the patch with the heart.
