AMG 397 is a novel inhibitor of MCL-1 under investigation for the treatment of multiple myeloma and acute myeloid leukemia. To ensure clinical supply of the drug substance, optimization of the reaction conditions and subsequent isolation to deliver a key advanced intermediate was required.  Commercial process development efforts resulted in improved reaction conditions, delivering consistent yields of the desired product.  Additionally, a unique multi-phasic crystallization was developed, which facilitated the filtration/isolation processes by producing material with superior particle properties. These improved process conditions were successfully demonstrated on manufacturing scale.
