The rapid and high-yielding synthesis of radiopharmaceuticals is greatly hampered by common issues with generator breakthrough and large elution volumes. Conventional in-solution radiolabeling from highly-diluted cyclotron eluates can yield low radiochemical yields and moderate purity. These issues necessitate secondary purification and concentration procedures that are tedious and incompatible with the short half-life of many radioisotopes.<br/>We will present approaches to the selective capture and release of radioactive isotopes from complex mixtures that efficiently address these aforementioned shortcomings. Specifically, we have devised a solid-phase based radiolabeling method that leverages pH dependence of complex speciation for the selective chelation of copper and gallium isotopes, followed by photochemical release of the desired radiolabeled product. We have validated our selective capture method by synthesizing two clinically translated radiopharmaceuticals and will discuss optimization of cargo-release efficiency, in the dependence of reaction volume, radioactivity and non-radioactive carrier.
