Automated high-throughput experimentation (HTE) is a powerful tool to combat the increasing complexity of pharmaceutical targets, the acceleration of clinical timelines, and the limited resources faced within the pharmaceutical industry. At Amgen, we have evolved our HTE capabilities to address the challenges of our differentiated pipeline and developed innovative chemistry that address the regioselectivity and stereoselectivity challenges arising from more complex molecular targets. In leveraging our HTE capabilities, we have been able to rapidly identify a novel Fe-mediated [<i>3+3</i>] cyclization to form tri-or tetra-substituted pyridines, discovered an unprecedented regioselective Pd-catalyzed cyanation, and developed catalysts for branched-selective Heck couplings.
