Bacteria produce sialic acid-like nonulosonic acids as part of their capsular polysaccharides (CPSs) and lipopolysaccharides (LPSs) which are bacterial virulence factors and potential vaccine candidates. These carbohydrates are attractive synthetic targets but pose significant synthetic challenges. One of such bacterium-specific nonulosonic acids is legionaminic acid. We have identified glycosyltransferases and related biosynthetic enzymes and have developed efficient chemoenzymatic strategies for synthesizing legionaminic acid-containing glycans. Furthermore, a fluorescent magnetic bead-based multiplex neoglycoprotein array system that we developed recently has been used to study glycan-protein interactions that are involved in microbe infection of hosts. Our recently results will be presented and discussed.
