One carbon-metabolism is central to several nucleotide, amino acid, and neurotransmitter biosynthetic pathways, and to epigenetic regulation. Dysregulation of these pathways supports metabolic rewiring in cancer and is associated with multiple neurological, cardiovascular, and diabetic diseases. The Chang lab at UC Berkeley is investigating one-carbon metabolic pathways via activity-based sensing of one-carbon metabolites, including formaldehyde (FA) and formate. My research in this area involves 1) investigation of the molecular basis of different FA release rates by the RNA eraser enzymes, FTO and ALKBH5, and 2) development of an activity-based sensing method for cellular formate using iridium transfer hydrogenation catalysis (ongoing). Our studies contribute to understanding of the roles which these endogeneous metabolites and biosynthetic precursors play in health and disease.
