The plant abscisic acid (ABA) sensing system which functions through a naturally occurring chemically-induced dimerization (CID) between PYR1 and protein phosphatase provides an appealing platform for the detection of small molecules. In this system, the ligand recognition is dependent on PYR1 which will simplify the new system development while the phosphatase could reduce the target off-rates and enhance the binding affinity. This platform has been successfully evolved up to 21 sensors with nM to uM sensitivities via engineering the PYR1. Even though this system could be employed in ELISA-like assays, the limit of detection (LoD) is still at the nanomolar range and the whole assay requires a relatively long time around one day to finish the detection. Herein, we developed a new detection platform by coupling the ABA sensing system and the Au-modified magnetic nanoparticles (Au-MPs) together. The MPs could facilitate rapid target capture and shorten the target recognition time. The Au on the MPs will magnify the obtained signal via plasmon enhance fluorescence (MEF) to increase the detection sensitivity. Finally, a highly sensitive and rapid small molecule detection platform is expected to be achieved.
