ICA-1 is a carbocyclic nucleotide that has been shown to display potent (IC<sub>50</sub> = 100 nM) and selective inhibition of the atypical protein kinase C iota (PKC-i) isoform. Its potential use has been explored in a variety of indications ranging from cancer to diabetes. Surprisingly, the enantiospecific synthesis of this compound has not been disclosed beyond its initial publication. We wish to report here our efforts toward a scalable and efficient route to the synthesis of not only ICA-1, but also to novel analogs in an effort to improve upon both its bioactivity and its physicochemical/pharmacological properties.
