Skin wounds in some patients can become a chronic illness imposing a risk of infection. Avoiding wound sepsis during an infection is highly important to prevent a fatal outcome from septic shock. However, it is a challenge to prevent infection with the emergence of antibiotic-resistant bacterial strains. Therefore, there is a need to develop wound care strategies with effective wound healing therapeutics with multi-functional properties. Over the years, Manganese (Mn)-based nanoparticles (MnNPs) have attracted immense interest as an antimicrobial agent. Despite their efficacy, nanotoxicity concerns due to their unpredictable fate in physiological conditions would likely face tough regulatory challenges. As an alternative, we have developed novel water-soluble chitosan vesicles loaded with bioactive N-acetylcysteine and Mn ions (Mn-C-N). Hydrothermally synthesized Mn-C-N vesicles were characterized using Scanning Electron Microscopy (SEM), Fourier Transform Infrared spectroscopy (FTIR), Energy-dispersive X-ray spectroscopy (EDS), among others. Antioxidant activity was evaluated with a scavenging assay, exhibiting an increase in activity over time. In-vitro antimicrobial activity against E. coli K-12 showed outstanding performance. The Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values were 16ppm of Mn. In-vitro cell viability was quantified for Human Dermal Fibroblast (HDF) and murine macrophage (J774a.1) to elucidate the biocompatibility and cytotoxicity. Overall safety profiles of Mn-C-N were better than MnNPs, suggesting strong potential for biomedical applications.
