Peroxisome proliferator-activated receptors (PPARs) belong to the nuclear receptors superfamily and are involved in lipid and glucose homeostasis, insulin sensitivity, and energetic metabolism. Therefore, these receptors represent valuable therapeutic targets for the treatment of metabolic diseases. The development of well-balanced PPARs modulators, devoid of the side-effects showed by the currently marketed PPARγ full agonists, is considered a major challenge. Herein we report a chemoinformatics search that let us identify a novel PPAR pan-agonist with a very attractive activity profile being able to reduce lipid accumulation and improve insulin sensitivity. Moreover, a strong correlation was found between stereochemistry and biological activity. This compound represents, therefore, a potential lead of a new class of drugs for the treatment of dyslipidemic type 2 diabetes.
