RIBOTACs are chimeric small molecules that eliminate disease-causing RNAs from cells through proximity-induced degradation. These molecules consist of a selective RNA-binding small molecule linked to a RNase L recruiting module that activates the endogenous ribonuclease to cleave the bound RNA. Since RNA structure has been implicated in diseases ranging from cancer to neurodegeneration, RIBOTACs offer an attractive method to reduce these RNAs and thus alleviate disease pathology. This work focuses on expanding current RIBOTAC technology to enhance the control chemical biologist have over cellular RNA degradation.
