Single-molecule FRET (smFRET) is one of the most popular and versatile methods to probe conformations and dynamics at the molecular scale. We show that by combining smFRET with Anti-Brownian Electrokinetic (ABEL) trapping, ultrahigh, shot-noise limited resolution of smFRET efficiency can be obtained together with single-molecule size and charge information. This multi-parameter approach expands single-molecules views of biological processes. We illustrate with examples in DNA-protein interactions, ribonucleoprotein assembly and peptide phosphorylation.
