The total synthesis of natural product analogs is a powerful method for accessing chemical probes that can be used to ask questions about biological systems. One of the key issues that arises in achieving interactively work between total synthesis and biological activity is how molecules can be evaluated quickly and accurately for their biological activity. In our group, we have been working to develop microelectrode array technology in order to address this issue. Microelectrode arrays contain thousands of individually addressable electrodes that can be used to monitor the current associated with redox active species in solution. The changes in current associated with the binding event can be monitored in real-time without the need to label the receptor or the molecule being studied. The Moeller group has been working with Ken Blumer lab at the WUSTL School of Medicine to understand the selective inhibition of YM-254890/FR-900359 to Gq protein. The arrays can potentially help in such an effort by providing rapid binding data that supplements more involved biological assays and can be used iteratively with synthetic efforts.
