Reactive oxygen species (ROS) are produced in small quantities during many physiological processes and include hydrogen peroxide (H2O2), superoxide (O2-), and hydroxyl radical (&lt;font size="1"&gt;.&lt;/font&gt;OH). Low concentrations of ROS are essential for the regulation of diverse physiological processes, such as immune cell activation and metabolic adaption. However, high concentrations of ROS oxidatively degrade biomolecules and cause oxidative stress, which is associated with cardiovascular, neurological, and inflammatory disorders. The deleterious impacts of ROS overproduction have spurred comprehensive efforts to develop strategies to control their in vivo concentrations, including the development of small molecule mimics of superoxide dismutase (SOD) and catalase (CAT). Here, we have prepared Mn(II), Fe(II), and Zn(II) complexes with the redox-active ligand 1,8-bis(2,5-dihydroxybenzyl)-1,4,8,11-tetraazacyclotetradecane (H4qp4) and characterized their abilities to mimic SOD and CAT. The Mn(II) and Fe(II) complexes were found to be SOD mimics as assessed by the hypoxanthine/xanthine oxidase/lucigenin assay, with IC50 values of 15.07 nM and 20.67 nM, respectively. CAT activity was identified for all three complexes by monitoring O2 production over time using a Clark-type O2-sensitive electrode, with kcat rate constants ranging from 2.6 × 103 s-1 to 3.7 × 103 s-1.
