Per- and polyfluoroalkyl substances (PFAS) are present globally in water, air, food and food containment, consumer products and dust. Children in communities with PFAS contamination sources may be particularly vulnerable to their health effects, as the chemicals are transferred to the infant across the placenta and in breast milk or contaminated water used to make formula. Recent studies have found enhanced metabolic disease in male and liver disease in female offspring of mice exposed to perfluorooctanoic acid (PFOA) and its replacement, GenX. Even with a far shorter half-life than PFOA, GenX proved to be a potent hepatotoxicant. Current work to determine subsets of genes highly associated with dose-dependent disease outcomes such as hepatocellular hypertrophy in exposed dams may help identify predictive biomarkers for use in testing other “data poor” PFAS.
