The application of high throughput chemistry to accelerate drug discovery programs and shorten the Design-Make-Test-Analyze (DMTA) cycle is of high importance to the pharmaceutical industry. This talk will summarize the technologies within the global Janssen Chemistry Capabilities, Analytical and Purification group that have been deployed to address bottlenecks in the cycle. The end-to-end parallel medicinal chemistry workflow is routinely applied to solve synthetic problems and enable rapid SAR exploration, addressing both target potency and ADME issues. The workflow encompasses: evaluation of reaction robustness, scaffold availability, building block diversity, final target selection, purification, and assay submission. Several vignettes will be presented to highlight how automation, library synthesis, and high throughput experimentation have culminated in the reduction of the DMTA cycle. Training of fellow team members in the application of the end-to-end workflow has led to high adoption and overall increased productivity within the department.
