Phytopathogenic fungi can cause plant diseases that reduce crop yield and quality. Mitochondrial complex III is one of the most promising targets for a number of pharmaceuticals and fungicides. Due to the wide-spread use of complex III-inhibiting fungicides, a considerable increase of resistance has occurred worldwide. Therefore, inhibitors with novel scaffolds and potent activity against complex III are still in great demand. Quinone inside inhibitors bind within the fungicidal binding site of the cytochrome <i>bc<sub>1</sub> </i>demonstrating broad spectrum fungicide disease control which are not cross resistant to quinone outside inhibitors. Inatreq is the first member of the QiI class of picolinamide fungicides to originate from research on the natural products antimycin A and UK-2A. This review highlights the emerging field of Qi inhibitors and discusses the bioisosteric principles and scaffold-hopping approaches employed in the discovery of this innovative class of fungicides.
