Multicomponent reactions featuring asymmetric catalysis have been widely applied in the synthesis of natural products, agrochemicals, and pharmaceuticals. The use of copper catalysts can allow for the borylative multi-functionalization of unsaturated carbon–carbon bonds, thus forging a key carbon-boron bond for use in further transformations. Additionally, isocyanates are readily available reagents that have recently attracted attention for the construction of new amide bonds. However, reports employing these versatile reagents for the enantioselective synthesis of amides are limited. We have recently developed a copper-catalyzed enantioselective amidation of allenes for the synthesis of borylated alkenyl amides with an adjacent stereogenic center. This process offers a new route to access these compounds with good yields, exclusive γ-regioselectivity, and high enantioselectivity. The reaction strategy and scope will be discussed.
