Potassium acyltrifluoroborates (KATs) are an important class of compounds with immense applications in bioconjugation, polymer, and materials chemistry. Recently, a copper-catalyzed carbonylative coupling strategy has been employed for the synthesis of aliphatic KATs that requires a very sophisticated reaction setup. This work represents a straightforward synthesis of such KATs utilizing a coupling between the mixed anhydrides of carboxylic acid and B2(pin)2 in presence of a commercial copper catalyst and subsequent quenching by aqueous KHF2. This method renders a broad substrate scope and is equally compatible with various aromatic substrates. Amino acid KATs are very important because of their role in peptide synthesis and protein conjugation using KAT ligation. This method provides a facile synthetic route for corresponding KATs in high yield, albeit the enantiopurity was slightly compromised. Future direction involves some modifications to ensure better enantiopurity for amino acid-derived KATs and addressing any existing limitations of the current method.
