Recent hardware and chemical developments utilized for automated glycan assembly (AGA) have expanded the possibilities of glycans achievable including those as large as 151-mers. Recent introductions such as a microwave generator allows for facile temperature manipulations from high temperature (100 °C) to low temperature (-40 °C) in minutes. This new addition helps to significantly shorten the time required for the incorporation of each building block, expands the portfolio of orthogonal temporary protecting groups, and aids in the synthesis of glycopeptides. Moreover, new protocols have been developed for the expedited synthesis of complex N-glycans and oligosaccharides containing either an anionic phosphate or sulfate moiety, <i>cis</i>-linkage, stapled cyclization, fluorescent probes, and furanosides. In total, these new tools allow for rapid access to homogeneous glycans, which are essential for medical, biological, and material science investigations.
