Photodynamic therapy (PDT) is an approach to treating cancer that is not widely utilized despite its ability to selectively target tumors over healthy tissue. PDT applies employs a photosensitizer (PS) a prodrug that uses light and oxygen to initiate localized cell death as well as an antitumor immune response. Historically, organic tetrapyrroles have been uses as PSs for PDT. Our group is specifically interested in metal-based PSs due to their tunability and often enhanced anti-cancer activity. This permits a tumor-centered approach that tailors the optical and photobiological properties of the PS to the specific type of cancer. Ruthenium- and osmium-centered PSs are particularly promising, with our own TLD1433 in phase 2 clinical trials for the treatment of non-muscle invasive bladder cancer. This presentation will focus on the design, synthesis, and characterization of a series of next-generation metal-based PSs.
