The synthesis of several cyclopropyl fluorescein esters was accomplished via a divergent synthetic strategy and contributed to a library of fluorescein esters. Bis(chloromethyl) fluorescein ether was used as a common starting material which was then combined via nucleophilic displacement reactions with a variety of carboxylic acids. Sixteen new cyclopropyl esters were produced and characterized by 1H-NMR, 13C-NMR, and HRMS. These fluorogenic esters can be employed to study enzymatic esterase activity. The new derivates were used in the biochemical assay of the BS2 serine hydrolase from Bacillis subtilis.
