One of well-known serious infectious diseases is anthrax, caused by gram-positive bacteria Bacillus anthracis. The vegetative cell wall polysaccharides of Bacillus anthracis is a potential carbohydrate antigen for the development of carbohydrate-based antibacterial vaccines for preventing bacterial infections. The hexasaccharide repeating unit of Bacillus anthracis vegetative cell walls contains several 1,2-cis glycosidic linkages including a 2-amino-2-deoxy-β-D-mannoside residue. Recently, our group developed an approach for stereoselective construction of β-mannosides and β-mannosamines via Cs2CO3 mediated anomeric O-alkylation. In this presentation, we will describe our efforts in the synthesis of a hexasaccharide repeating unit of Bacillus anthracis vegetative cell walls using our recently developed methodology.
