The reactivity of inorganic nanoparticles may vary in an organic medium according to their aggregation states and corona formation. Recent studies showed that these characteristics may have an important effect on the toxicity of nanoparticles. In this study, we aimed to examine how the interaction with a real food matrix affects the in-vitro toxicity of TiO2 nanoparticles. First, TiO2 nanoparticles (25nm, 100nm, and 250 nm) were incubated with the whole milk and digested with in-vitro simulation solutions. After digestion, aggregation states and corona structure of nanoparticles were studied with electron microscopy, DLS, and zeta potential. The human colon adenocarcinoma cell line Caco-2 was used for the investigation of in-vitro toxicity. The effect of digested TiO2 NPs on cellular viability was studied by using 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide (MTT) assay and the level of reactive oxygen species (ROS) was measured by using 2’,7’-dichlorofluorescin diacetate (DCFH-DA). In conclusion, the in vitro toxicity of nanoparticles in a real food sample after digestion was assessed in detail. These outputs will be shed light on the behavior of common additives in food matrix aggregation situations and protein corona formation.
