The implication of glycans in a wide range of fundamental and aberrant biological pathways has created a demand for diverse molecular probes for elucidation of these mechanisms. Carbohydrate-like structures that are structurally similar to naturally occurring glycan mediators (broadly termed “glycomimetics), are particularly sought after, as they are likely to mimic, promote or antagonize the activity of their parent molecules. Invariably such structures are not naturally occurring and often require chemical synthesis. However, their intrinsic complexity and diversity presents a challenge to the development of general synthetic methods. In this context we have been interested in syntheses of tailored carbohydrate-like molecules that encompass the chemistry of n-pentenyl acetals and glycosyl crotylating agents. The evolution of these methodologies, and the synthesis and applications of glycomimetics therefrom will be presented.
