Tumor-associated carbohydrate antigens (TACAs) are glycan stuctures that are modified relative to those on normal cells and play important roles in cancer proliferation and aggression. They are called "antigens" since, although still self-molecules, the immune system mounts a response to these glycan aberrations and hence many TACAs have been targets of antitumor vaccine design. In additon, TACAs can be recognized by specific tumor-associated lectins leading to enhanced tumor progression and metastasis. We have been interested in one particular TACA, the Thomsen Friedenreich antigen (TFag) disaccharide, as a basis for both antiadhesion and tumor vaccine therapies through the development of glyconanomaterials containing the TFag in a variety of presentations. This talk will cover our recent work in this area, while also relating the influence that Professor Fraser-Reid had the direction of this research.
