Cereblon E3 ligase modulating drugs (CELMoDs) are an important class of oncology treatments. The structures of these compounds feature an isoindolinone core wherein the nitrogen at the 2-position is incorporated into an a-glutarimide moiety. CC-90009 and CC-220 (Iberdomide) are two CelMODs currently in clinical trials for the treatment of Acute Myeloid Leukemia and Multiple Myeloma, respectively. The syntheses of these compounds used to support clinical trials will be discussed, including route development and challenges associated with scale up. The requirement to control chirality of the a-glutarimide moiety highlights a difference in the approaches, while a mechanistic detail of the isoindolinone construction underscores a common theme.
