Purine-derived compounds are widely investigated as cyclin-dependent kinase inhibitors in the design of neuroprotectors. The neuroprotection of the purine-based kinase inhibitors, on the other hand, maybe multifactorial, including their role as modulators of oxidative stress. However, the role of the kinase inhibitors as oxidative stress modulators has not been established to date. Toward developing purine-based neuroprotectors that can modulate oxidative stress, we have synthesized a series of purine-based ATP-analogs and investigated their oxidative stress effects using SHSY-3Y human neuroblastoma cell line and MTT assay for cell viability. Through these studies, we have identified purine-based neuroprotector compounds that favorably modulate oxidative stress induced by the Fenton reaction-generated reactive oxygen species (ROS).
