Acute Lung Injury is characterized as an innate immune system response that can develop into a major clinical syndrome such as ARDS. Common pathophysiologic features of acute respiratory distress syndrome (ALI/ARDS) include pulmonary neutrophils infiltration, interstitial edema, and impaired gas exchange. such as HNE /PR3 and MMP-2 are playing an essential role in mild ARDS. Therefore, there is an urgent need for the development of a rapid biosensing platform for the detection of ARDS. The aim of the present study is to develop a paper-based multiplexing sensing platform for the detection of ALI/ARDS in infected mice after intranasal administration of LPS. Colorimetric paper-based sensor as a base for substrate sensing was developed. Multiplexing of PR3, MMP-2 analyzes resulted in high peptide cleavage after 30 min, 2, 4, and 24 hours of LPS administration. Multiplexing platform of three analytes Elastase, PR3 and MMP-2 in a relatively high peptide cleavage after 30 min and 24 h only These data indicate the potential of neutrophils to induce the release of serine protease peptides within the first 30 min of LPS administration. The results demonstrated that both MMP-2 biosensors and the multiplexing platform of PR3 and MMP-2, can provide a promising biosensing platform for ALI/ARDS in infected mice at all stages (30 min to 24h), while the multiplexing platform of Elastase/MMP-2 can be useful for early (30 min) and late stages (24 hrs) of ALI/ARDS. Further studies needed to apply these potential diagnostic biosensing platforms on ARDS in patients.
