Melanoma is one of the most dangerous cancers and it is the deadliest form of skin cancer accounting for 75% of skin cancer deaths. Natural products are the continuous sources of novel drug discovery due to a wide range of bioactive compounds and are particularly useful as anticancer therapeutics. According to a recent analysis, it has found that around 50% of anticancer drugs approved from 1940 to 2014 were either originated from natural products or derived directly from them. Verbena officinalis (Verbenaceae) is commonly used in traditional medicines of different countries. Therapeutic studies have demonstrated its important bioactivities including anti-inflammatory, antioxidant, anticonvulsant, antimicrobial, anticancer, neuroprotective, and analgesic. In this study, the methanolic extract of Verbena officinalis (whole plant) and its different soluble fractions (n-hexane, chloroform, ethyl acetate, n-butanol, and acetone) with increasing polarity order were prepared and evaluated for the in vitro anti-melanoma potential employing B16 melanoma cell line. Then n-hexane, ethyl acetate, and n-butanol fractions were subjected to column chromatography (normal phase silica gel, wet column packing) by using different solvent systems. Preliminary results exhibited that some sub-fractions as well as pure compounds were the most active against melanoma B16 cell line and need to be characterized by NMR spectroscopy and Mass spectrometry. These findings suggest that Verbena officinalis could be further investigated as a promising candidate to treat melanoma in the future.
