Despite significant advances in synthetic strategies aimed at functionalizing alpha and beta positions of enones or carbonyl groups, functionalizing their gamma position remains challenging. However, issues regarding the selective activation of these groups at the gamma position relative to carbonyl have always been in the interest of the synthetic community due to their potential scope to access new structures in a more efficient approach. <br/>1,4-Amino ketone moieties are prevalent in many natural products biologically active pharmaceutical drugs and are used as a building block in many organic syntheses. We report the synthesis of 1,4-amino ketone derivatives using Cu (I) catalyzed regioselective gamma C-N bond formation of enones. This can be achieved via Nitroso Diels alder reaction of phosphate dienol ethers and nitroso benzene followed by N-O bond cleavage. The range of available strategies to access the gamma C-N bond relative to carbonyl is limited. This methodology will overcome limitations and provide various gamma C-N carbonyl-bearing bioactive molecules.
