Multicomponent coupling reactions (MCRs), such as the Ugi-Smiles reaction, provide direct access to diverse scaffolds from simple starting materials. In the presence of methanol and a conjugated aldehyde, a novel five-component oxa-Michael Ugi-Smiles reaction was observed. A broad range of substituted alcohols and isocyanides were successful, with moderate, but promising, yields (40-70%). Current efforts are focused on variation of the amine and aldehyde components, as well as exploring the reaction pathway through competitive studies with β-methoxy substituted aldehydes.
