UHRF1 is a multi-domain epigenetic protein that plays an essential role in regulating gene expression by binding histones and methylated DNA. The binding of UHRF1 to histone H3 tri-methylated at lysine 9 (H3K9me3) is well characterized, however, we have recently discovered that UHRF1 also interacts with a novel histone modification. To better understand the importance of this interaction, we have utilized pull-down assays and Western blots to show whether this interaction occurs in cells.
