Breast cancer is the second most common cause of cancer related deaths in women. Over 40,000 women die from breast cancer each year just in the United States. There are three types of breast cancer based on the over expression of specific receptors. Most Breast cancer patients show expression of an estrogen receptor (ER+), progesterone receptor (PR+) and human epidermal growth factor 2 (HER2+). Breast cancer with expression of all three receptors is referred to as triple positive breast cancer. The absence of over expression of these three receptors is termed as the triple negative breast cancer (TNBC). p70S6Kinase 1 (S6K1) is a member of the AGC (cyclic AMP-dependent protein kinase, cyclic GMP-dependent protein kinase, and protein kinase C) subfamily of serine/threonine protein kinases. S6K1 is the downstream target of mTOR in the PAM pathway. This pathway is involved in the regulation of various physiological and cellular processes in breast cancer. Our research group identified 6-amido-4-aminoisoindoline-1,3-diones as S6K1 inhibitors. Docking studies revealed that 6-amido-4-aminoisoindolin-1-ones could have better binding to the kinase. The research the design and synthesis of these molecules that can potentially inhibit the growth of breast cancer is the goal of the project. The synthetic scheme for the designed molecules is currently being established.
