Lysine methylation is a functionally diverse chromatin modification that regulates a number of cellular processes including heterochromatin formation, transcriptional regulation and DNA repair. The spatial and temporal control of histone methylation is achieved by opposing activities of histone modifying enzymes - methyltransferases, enzymes that deposit methylation marks, and demethylases, enzymes that remove them. Methylation states of chromatin are recognized by methylation reader domains. Misregulation of methylation is associated with disease, including cancer and neurodegeneration, and inhibitors of methylation regulators offer promising new avenue in oncology. Our recent work on development of cyclic peptide ligands for readers of chromatin methylation will be discussed.
