Cereibacter sphaeroides, formerly known as Rhodobacter sphaeroides, has a complete genome sequence with many proposed genes without a defined function. Genome anaylsis found several genes with proposed function including the gene with the gene symbol RSPH17029_RS05495 as described as a "circularly permuted type 2 ATP-grasp protein." BLAST analysis found the primary protein sequence shows sequence similarity to an enzyme that catalyzes the synthesis of the biomolecule glutathionyl-spermidine using glutatione, spermidine, and ATP as substrates. This gene has been amplified by PCR using the genomic DNA as a template. The resulting PCR product has been inserted into a pET expression vector that includes a His-tag and an IPTG inducable promotor. The protein has been expressed and purified. The kinetic parameters have been determined to be 0.114 mM for the Km with spermidine as a substrate. The kinetic parameters obtained thus far have been comparable to the published parameters found from other organisms.
