Antimicrobial resistance (AMR) has become a rising global health threat for human beings. To tackle the AMR problem, there are many approaches including mimicking the innate immune responses. Antimicrobial peptides (AMPs) are a first-line response to pathogens. They are amphiphilic biomacromolecules that are composed of tens of amino acids and cationic charges. However, AMPs and their synthesis have some drawbacks such as toxicity and high manufacturing cost. In this study, we have been inspired by the AMPs to synthesize therapeutic interventions to curb AMR pathogens. Facially amphiphilic bicyclic rings and multiple cationic charges were attached to each repeat unit of polymers to form membrane-active therapeutics. Facial amphiphilicity is an extraordinary feature of a variety of AMPs to battle pathogenic bacteria which reduces the local and global arrangement forfeit. The structure-activity relationship of facial amphiphiles on antimicrobial efficacy was investigated. We believe this approach will pave the way for discovering new antimicrobial agents.
