Carbohydrates are attractive biomolecules for use in the development of materials to interface with biology. This is because many biological interactions are dictated by carbohydrate recognition, from cellular adhesion and molecular recognition to the uptake of extracellular materials. Moreover, carbohydrates play significant roles in disease progression and in modulating immunity. The composition of carbohydrates, as well as how they are structurally arranged on scaffolds (e.g., proteins, lipids, extracellular matrices), dictates the function and biological role of glycopolymers and glycan conjugates. However, our understanding of the fundamental chemistry that underlies their recognition and higher order assembly remains incomplete. In the Callmann lab, we have initiated a program geared towards the development of chemical strategies to interrogate the relationship between carbohydrate structure and biological function, such that we can uncover key parameters governing carbohydrate- directed biological processes. This presentation outlines our efforts-to-date towards the development of a comprehensive understanding of how carbohydrate structure (as presented on polymeric scaffolds that closely mimic their native conformations) impacts their higher order assembly and biological recognition, using carbohydrate-polymer conjugates (CPCs).
