Accurately and precisely measuring lipoprotein subclasses is necessary for a multitude of applications downstream such as early detection of cardiovascular disease (CVD). Clinical methods incorporate labor and time intensive immunochromatographic techniques to quantify lipoprotein subclasses, and several methods use a derivative calculation to estimate lipoprotein fractions indirectly. During recent years, these methods alone show difficulties in achieving the precision required for distinguishing individuals with and without CVD. The ultimate goal of our research is to prove the feasibility of the rapid, single-spin fractionation of serum lipoproteins by adapting Density Gradient Ultracentrifugation (DGU) with Sucrose/NaCl solution and fluorescence detection techniques. Lipoprotein fractions are separated by density and isolated for further characterizations including fluorescence detection with NBD C6 Ceramide and DMSO. This method is direct, efficient, reproducible, facile, and rapid. DGU lipoprotein profiling is a powerful tool that can examine patients at risk of cardiovascular disease and pave the way for early detection and prevention of CVD.
