Triple-Negative Breast Cancer (TNBC) is a very aggressive and rare cancer affecting about 10% of breast cancer patients. African American premenopausal women who are younger than 40 years of age, as well as those with the BRCA 1 mutation, are more susceptible to getting TNBC. This specific type of cancer has negative expression of the estrogen receptor (ER), progesterone receptor (PR), or the human epidermal growth factor receptor 2 (HER-2). Estrogen and progesterone are both female hormones, while HER-2 is a protein. Chemotherapy is often used since other treatment options are ineffective due to the missing receptors. Naturally occurring compounds from plants, known as phytochemicals, serve as vital resources for novel drugs and are also sources for cancer therapy. The phytochemical berberine has recently been shown to inhibit tumors such as hepatoma, leukemia, breast, lung, colon, ovarian, and cervical cancer cells. Previous research has shown that the analogs formed on the C12 position of berberine will enhance the inhibition of breast cancer cells. Herein, we will exhibit the synthesis of aryl berberine analogs via the Suzuki Coupling reaction utilizing microwave chemistry. Each berberine analog will undergo chemical purification and analysis. The series of analogs will be evaluated for the ability to inhibit the growth cells superior to the parent compound berberine and its effect on targeting the <i>PALB2</i> gene.
