Selective oxidation of hydroxypropyl polysaccharides has been innovated in recent years and is perhaps the most efficient, selective route to ketone-substituted polysaccharides. We describe in this presentation how these ketone-terminated oligohydroxypropyl substituents can be exploited in many ways. We will describe how they may be used to create all-polysaccharide hydrogels, with considerable promise for controlled release, space-filling, lubrication, and other applications. We will describe how they can be used to link active molecules to the polysaccharide, either permanently or by labile linkages. We will describe new families of derivatives that can be synthesized in this way, including amino acid-polysaccharide constructs, polyzwitterionic derivatives, and derivatives with considerable promise for amorphous solid dispersion of drugs to enhance their solubility, and hence bioavailability.
