Innate immune cells detect pathogens through simultaneous stimulation of multiple receptors, but how cells use the receptor crosstalk to elicit context-appropriate responses is unclear. In this talk, I will highlightour recent studies on imaging the organization and crosstalk of receptors during innate immune activation. Inrecent studies, we have shown that various types of innate immune receptors form separate nanoclusters that interact at their interfaces to enable signaling crosstalk. Different from the prevailing view that immunereceptors colocalize to synergize their signaling, the crosstalk between many innate immune receptors occurs through interface interactions between non-colocalizing receptor nanoclusters. Their interactions and thereby signaling outcome are modulated by the context, including mobility of ligands and lateral spacing between ligands.
