A new synthesis method for the preparation of (S)-oxetan-2-ylmethanamine (1) was developed. Previous synthesis approaches used sodium azide, a hazardous material, to form an intermediate organoazide followed by reduction to introduce the amine nitrogen. Our method avoids the use of azide and organoazide intermediates and introduces the amine nitrogen through the use of dibenzylamine. This results in an improved process safety profile for the overall synthesis of 1. We will discuss the route development and optimization to provide synthetically useful amounts of oxetane 1.
