Metal-catalyzed couplings are frequently used as a means for the efficient assembly of pharmaceutical targets, especially using Pd-catalyzed methods. To support sustainability goals, methods using nonprecious metals such as Cu, Ni or Fe are needed to replace Pd methods for manufacturing on large scale. A family of 6-hydroxypicolinamides has been identified as effective supporting ligands for Cu-catalyzed C-N and C-O couplings of heteroaryl bromides and chlorides. The structure activity relationship of the ligands as well as the scope of the couplings will be discussed.
