The quantification of chlorophyll in leafy biomass is a common measurement method for various reasons including assessing plant health, determining the effect of growth conditions, or understanding the impact of foraging to name a few. While soil-plant analysis development (SPAD) meters are commonly used for such analysis, the degree to which SPAD measurements agree with and represent chlorophyll concentration across plant species is unclear. As a result, it would be advantageous to develop a method for chlorophyll extraction and quantification that can to carried out in remote field locations. To that end, several reported lab methods were combined and modified to allow for chlorophyll quantification using UV-Vis and fluorescence spectroscopy. Additionally, data was compared with average SPAD meter data to validate several method modifications for quantification of chlorophyll in buckthorn (<i>Rhamnus cathartica) </i>leaves specifically. Data from all three methods, UV-Vis, fluorescence, and SPAD meter, is reported, compared and discussed.
