Pyridinophane molecules have recently been shown to have both antioxidant and pharmacological properties suitable for therapeutic applications targeting neurodegenerative diseases including Alzheimer’s. However, blood brain barrier permeability has proven a difficult challenge to overcome. We have synthesized derivatives of the parent molecules with substitutions designed to enhance permeability and, potentially, increase the antioxidant activity beyond that of the parent molecules in hopes of producing a molecule suitable for pharmacological testing in animal models. These substitutions have been successfully achieved in two different positions of the pyridine scaffold: on the pyridine ring (<b>L1</b>), or as an appendage on the N-atom of the macrocycle (<b>L2</b>). The results presented here will focus on synthesis and evaluation of the substitution of the 8-hydroxyquinoline moiety and comparing its antioxidant activities to similar congeners to understand how the position of substitution controls reactivity.
